For substrate concentration \(S\), the Michaelis–Menten relationship is
\[v(S)=\frac{V_{\max}S}{K_m+S}.\]At \(S=K_m\), the reaction rate is half its maximum:
\[v=\frac{V_{\max}}2.\]Worked investigation
Choose values for \(V_{\max}\) and \(K_m\), calculate \(v(S)\) over a range of substrate concentrations and plot the saturation curve. Increasing \(V_{\max}\) changes the upper rate, while \(K_m\) changes the substrate scale at which saturation occurs.
Data extension
Given measured pairs \((S_i,v_i)\), estimate \(V_{\max}\) and \(K_m\) by nonlinear least squares and compare fitted predictions with observations.
